Schematic depiction of the maintenance of the reading frame for the translational initiation of non-formylated methionine Escherichia coli initiator tRNA (E. coli Met-tRNAi). Structure of single fluorescent methionine E. coli initiator tRNA (fluorescent Met-tRNAi), which initiates the expression of proteins of interest (enhanced green fluorescent protein or Tat). First, a non-formylated group was attached to the free amino group of Met-tRNAi. Met-tRNAi was fluorescent-labeled via a reaction with its amine reactive group. Methionylation via E. coli methionyl tRNA synthetase (EcMRS) charged E. coli Met-tRNAi for incorporation into the proteins of interest in HeLa cells
