DNA fragmentation and nuclear-morphology changes induced by CB01. A DNA fragmentation. U87 and HeLa cells were cultured with 10 and 40 nM CB01, respectively, in 60 mm culture plates. After 48 h, the cells were lysed, and the supernatants of the lysates were collected and subjected to phenol–chloroform extraction. DNA was harvested by ethanol-precipitation and then treated with RNase A for 2 h. The positive control for the DNA fragmentation was cells treated with 5 µM camptothecin; a 1 kb DNA size marker was used. B Evaluation of nuclear morphology.
